GALLY International Biomedical Research & Consulting LLC provides
expertise in light and electron microscopy to clients in both
academia and commercial industry worldwide. We will provide
assistance with the experimental design, microscopic analysis, and
manuscript and grant writing for both biological and non-biological
studies for the BS, MS as well as MD&PhD program and postdoctoral
students and scientist at any level and any other organization
looking for modern ultrastructural analysis.
Dr. Aliev has extensive expertise in all of the various aspects
of microscopic analysis such as experimental design, data analysis,
and manuscript/grant writing for both biological and non-biological
studies. With many years of experience in LM, EM, 2-photon
microscopy, Atomic Force and Confocal Microscopy he has produced
pioneering work in different areas of EM, including new methods of
EM techniques. These techniques include cytological in situ
hybridization at the light and electron microscopic levels using
non-isotopic colloidal gold probes, peroxidase-anti-peroxidase
(PAP), and pre- and post- embedding single double and triple
immunogold cytochemistry and quantification. His methods have been
included in studies published in a number of highly recognized
journals (for details, please see the list of publications in the
media portion of our site and pubmed search: aliev g).
GALLY International’s management team also continues an active
research program. One of it’s latest research interests focuses on
investigating the interaction of nanoparticles with tissues and
cells. Dr. Aliev has recently developed ways to determine the
biological effects of nanoparticles in vivo and in vitro by
utilizing animal models of human diseases. The goal of this study
is to not only elucidate the pathogenic mechanisms underlying the
nanoparticles' effects, but to also discover potential new drug
development strategies. For example, ongoing several projects
supervising by Professor Aliev (Strategies for Drug Development and
Delivering to Central Nervous System) has so far showed promising
results regarding the specific delivery of drugs to posttraumatic,
tumor and animal model that mimics human neurodegeneration. In
another of Professor Aliev in vivo studies, he had found that
nanoparticles are able to cross the blood brain barrier (BBB),
which has been the biggest impediment in delivering drugs to
patients with Posttraumatic Brain Injury as well as Alzheimer
disease. GALLY International’s teams of professionals are using
classical biochemistry, cell biology, and morphology techniques in
conjunction with more modern methods, such as SEM/TEM SED X-ray
elemental analysis and NMR studies of protein structure and
dynamics, macromolecular interactions, mechanistic enzymology, and
computational analyses and modeling, in order to determine the
exact nature of the relationship between the nanoparticles and the
underlying tissue.

Examples of work performed by the GALLY
International:
- Perfusion Fixation: The trained technical
staff at GALLY International can either assist or perform the
perfusion fixation of large and/or small laboratory animals as
well as cell culture or cell suspensions. Fresh solutions will be
prepared for each perfusion. The company also provides vibratome
block sectioning for future immuno or routine microscopic
investigations.
- Routine Light Microscope semithin sections (LM and EM):
Samples, including fresh tissue specimens, cultured cells on
substrate, subcellular fractions (for purity determinations) as
well as plastic or other organic and non-organic materials can be
routinely processed (paraformaldehyde/glutaraldehyde-osmium fixed,
dehydrated plastic embedded) and thick or ultrathin sections
obtained.
- Immunolocalization: Studies at both the light
and EM level can be performed using pre- and post-embedding
single, double, or triple immunogold and/or
peroxidase-anti-peroxidase (PAP) or other tracers to visualize
target molecules. Examples of Immunolocalization studies using
colloidal gold and PAP techniques at the light and EM levels can
be found in our recent publications.
- Cytological Localization of in situ hybridization
markers for DNA: Studies at both the light and EM levels
can be performed using PAP and colloidal gold (non-direct,
non-isotopic) probes to visualize the cytological localization of
DNA markers at the cellular and subcellular levels. We have found
that cytological localization of in situ hybridization markers for
wild and deleted mitochondrial DNA provide more correct
information about the mitochondria DNA deletions than that other
techniques, including PCR. (For details on in situ hybridization

- Rapid freezing of tissue samples can be used for
Immunolocalization of freeze-substitution.
- GALLY International can employ the techniques of either negative
staining or rotary metal shadowing of
individual proteins to allow high resolution imaging of the shapes
and dimensions of individual protein molecules or complexes.
- Using the automated computerized systems, high quality
immunofluorescent and PAP stains can be obtained using different
antibodies for different tissue or cell culture systems.
- Three dimensional structural organizations of the cells,
tissue, organs as well as intracellular compartments can be
performed by using Scanning Electron Microscopy. Our highly
trained staff can perform tissue processing, critical point
drying; sputter coating, microscope operation and imaging of
cells, tissues, and other organic or non-organic materials.
- Data Interpretation: The GALLY International
Biomedical Research & Consulting LLC also offers data analysis and
interpretation along and with assistance in manuscript and grant
